This MAb recognizes protein of 26 kDa-60 kDa, which is identified as CD63. Its epitope is different from that of MAb LAMP3/529. The tetraspanins are integral membrane proteins expressed on cell surface and granular membranes of hematopoietic cells and are components of multi-molecular complexes with specific integrins. The tetraspanin CD63 is a lysosomal membrane glycoprotein that translocates to the plasma membrane after platelet activation. CD63 is expressed on activated platelets, monocytes and macrophages, and is weakly expressed on granulocytes, T cell and B cells. It is located on the basophilic granule membranes and on the plasma membranes of lymphocytes and granulocytes. CD63 is a member of the TM4 superfamily of leukocyte glycoproteins that includes CD9, CD37 and CD53, which contain four transmembrane regions. Biotin-HPDP is a reagent widely used to introduce a biotin moiety to a sulfhydryl group of protein via a reversible disulfide bond via the Biotin switch method. This reagent is useful for affinity-purification of biotinylated proteins by using avidin-coated beads, because the disulfide bond can be cleaved by a reducing agent. However Biotin-HPDP has very low solubility in a solvent, and it is necessary to dissolve it by using an organic solvent such as DMSO or DMF. The preparation of this solution is time-consuming.Biotin-HPDP(WS) solution is a novel water-soluble biotin-labeling reagent that overcomes the problems of Biotin-HPDP by dramatically improving its solubility in water. CF Dye Succinimidyl Esters (SE, also known as NHS esters) are amine-reactive fluorescent dyes. SE dyes are commonly used to label antibodies and other proteins on free amine groups, such as lysine residues. CF dyes are Biotium's line of next-generation fluorescent dyes with advantages in brightness, photostability, and conjugate specificity compared to other fluorescent dyes. Biotium Fura-2, AM Ester, 20x50ug
Fura-2, AM ester is an intracellular fluorescent calcium indicator, and the preferred dye for ratiometric imaging microscopy. The membrane-permeant AM ester can be loaded into cells via incubation, but due to limited water solubility, a mild detergent Pluronic F-127 is often used to facilitate cell loading. Fura-2, AM ester itself does not bind Ca2, but intracellular esterases readily hydrolyze it to Fura-2. Upon calcium binding, the excitation maximum of Fura-2 undergoes a blue-shift from 363 nm (no Ca2) to 335 nm (high Ca2) while the emission maximum is relatively unchanged. Measure by flow cytometry, fluorescence microscopy, or certain fluoresence microplate readers. Recognizes a 67 kDa glycoprotein, which is identified as CD33. It is a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It belongs to the immunoreceptor tyrosine-based inhibitory motif (ITIM)-containing molecules able of recruiting protein tyrosine phosphatases SHP-1 and SHP-2 to signal assemblies; these ITIMs are also used for ubiquitin-mediated removal of the receptor from the cell surface. CD33 is expressed on cells of myelomonocytic lineage, binds sialic acid residues in N- and O-glycans on cell surfaces, and is a therapeutic target for acute myeloid leukemia. CD33 is expressed on myeloid progenitors, monocytes, granulocytes, dendritic cells and mast cells. MAb B-R18 specifically recognizes CD95, also known as Fas, a transmembrane glycoprotein with a MW of 40-45 kDa, containing 8 kDa of N-glycosidic-linked polysaccharide. It is a receptor for TNFSF6/FASLG, a member of the nerve growth factor receptor/tumor necrosis factor superfamily, mediating receptor-triggered apoptosis. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation, which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis.
